This dual action increases serotonin activity in the central nervous system, contributing to its antidepressant effects.

Pharmacokinetics Overview

Trazodone has a variable absorption rate with peak plasma concentrations occurring 1 to 2 hours after ingestion. It has a large volume of distribution, indicating extensive tissue binding. Metabolism primarily occurs in the liver through the CYP3A4 enzyme, producing an active metabolite m-chlorophenylpiperazine (mCPP). The elimination half-life ranges from 5 to 13 hours depending on dosage.

Indications and Usage

Trazodone is indicated for the treatment of major depressive disorder (MDD). It is also prescribed off-label for insomnia due to its sedative properties. Additionally, Trazodone is used in managing anxiety and certain pain syndromes.

Dosage and Administration

Special Populations Considerations

Elderly patients may be more sensitive to the sedative effects of Trazodone and may require lower initial doses.
